ABSTRACT

Objective:Meta analysis was used to evaluate the diagnostic value of Septin-9 methylation in colorectal cancer.Methods:Research on the diagnostic value of Septin-9 methylation in WanFang, CNKI, Weip, Pubmed and Embase. The release time is from inception to January, 2020. After data extraction, QUADAS-2 standard was used to evaluate the quality of the included literature. Then the extracted data were preliminarily combined to determine the threshold effect, heterogeneity and diagnostic efficacy, and the source of heterogeneity and subgroup analysis were further discussed through sensitivity analysis. Finally, funnel plot was used to evaluate whether publication bias existed.Results:A total of 17 articles were included in the study, including 1 668 cases in the case group and 2 407 cases in the control group.The combined sensitivity: 0.74 (95% CI: 0.72-0.76), specificity: 0.81 (95% CI: 0.79-0.83), positive likelihood ratio: 6.89 (95% CI: 3.39-12.19), negative likelihood ratio: 0.30 (95% CI: 0.23-0.40), diagnostic ratio: 23.43 (95% CI: 10.25-53.53), area under curve (AUC) of SROC (summary ROC): 0.821. Subgroup analysis showed that Septin-9 methylation detection was of high diagnostic value in the diagnosis of colorectal cancer by tissue samples and nested methylation specific PCR (DOR=63.355, 56.567), while the diagnostic value after surgical intervention is low (DOR=14.245). Conclusions:The methylation detection of septin-9 gene in Chinese population is of high value in the diagnosis of colorectal cancer and is expected to become a diagnostic marker of colorectal cancer.
